One step transformation of Yarrowia lipolytica

1. Single colony 16-24hr 28C culture on YPD plate.
2. Cell, about 5 x 10^7 cells,을 tip으로 one step buffer 100uL에 넣고 vortexing to resuspend.
3. Add 1uL, 1ug 이상, of vector or DNA fragment and vortex.
4. 강하게 tapping 하면서 inverting 하는 것이 vortexing보다 권장된다.
5. RT incubation 30min.
6. 39C 1hr incubation for heat shock.
7. RT incubation 10min to recovery.
8. 4C centrifuge 8000rpm 1min.
9. 이 때 pellet이 혜성 꼬리처럼 흘러야 잘 형성된 것이다.
10. Discard supernatant.
11. Add YPD 800uL and pipette.
12. 28C 2hr incubation to cell recovery.
13. 4C centrifuge 8000rpm 1min.
14. Discard supernatant.
15. Add pure water 100uL and resuspend cells.
16. Spread mixture on selective media plate.
17. Selection media plate는 antibiotics plate 혹은 auxotrophic plate를 사용한다.
18. Volume을 늘릴 필요가 있을 때는 one step buffer나 YPD media를 사용한다.
19. Incubate at 28C for 36-48hr.
